ABSTRACT

Objective To compare and analyze biomechanical stability of Ethibond thread suture and threaded anchor for fixing inferior pole fracture of the patella using the finite element method. Methods CT images of the knee joint from a healthy adult volunteer were selected and the inferior patella fracture model was established, then the three-dimensional (3D) models of fracture fixation with Ethibond thread suture and threaded anchor were established. Stress and displacement distributions of each fixiation model at different knee flextion angles were analyzed. Results In the range of 0°-90°, as knee flexion angle increased, the maximum stress and maximum displacement also increased. At the same knee flexion angle, threaded anchor fixation had the highest stress level, and Ethibond thread suture had the lowest stress level and the most uniform stress distributions. The displacement deformation of Ethibond thread suture was the largest, and the displacement deformation of threaded anchor fixation was the smallest. Conclusions The maximum displacement of the seam suture around patellar tendon with Ethibond suture is relatively moderate, and its maximum stress is relatively small, which is a reliable method for fixing inferior pole fracture of the patella in clinic.

ABSTRACT

Osteoporosis is a chronic skeletal disease characterized by low bone mass, destruction of bone tissue microarchitecture, and imbalance of bone homeostasis, leading to increased bone fragility and increased risk of fractures. Oxidative stress caused by the disruption of the balance between excess reactive oxygen species (ROS) and the anti-oxidative system is an important factor in the occurrence and progression of osteoporosis. Nuclear factor E2-related factor 2 (Nrf2)/heme oxygenase-1 (HO-1) is an important anti-oxidative stress pathway. Nrf2 is a primary factor in regulating cellular oxidative stress. Activating Nrf2 can stimulate the expression of HO-1. HO-1 is a key enzyme whose metabolites are bile green Oxygen, carbon monoxide, and free iron. The metabolites can scavenge ROS, thereby exerting an antioxidant effect in cells. At present, domestic and foreign scholars have reported that the Nrf2/HO-1 signaling pathway is closely related to the occurrence and development of osteoporosis and the mechanism of drugs. Chinese medicine can effectively solve the insufficiency of western medicine with multi-target, multi-channel, and multi-level advantages. Chinese medicine can resist oxidative stress, inflammatory response, and apoptosis by regulating the Nrf2/HO-1 signaling pathway, thus treating osteoporosis. This article reviewed the relationship between Nrf2/HO-1 signaling pathway and its key target protein factors and osteoporosis, to clarify the important role of the Nrf2/HO-1 signaling pathway in osteoporosis. At the same time, a systematic summary of Chinese medicines targeting and regulating the Nrf2/HO-1 signaling pathway for the treatment of osteoporosis was conducted, to provide a theoretical basis for further precise treatment of osteoporosis.

ABSTRACT

Objective:To investigate whether SKA1 is a key molecule regulating malignant proliferation of liver cancer, and further explore its mechanism to provide molecular theoretical basis for subsequent targeted therapy. Methods:The data of liver cancer from TCGA database were analyzed by bioinformatics technology. The expression of SKA1 in liver cancer was analyzed. At the same time, we also analyzed the relationship between the expression of SKA1 and the prognosis of patients with liver cancer. The hepatoma cell line overexpressing SKA1 was constructed by liposome-mediated cell transfection technique, and the effect of SKA1 on the proliferation of hepatoma cells was further tested by CCK-8 and plate cloning assay. At the same time, we found that E2F1 is also highly expressed in liver cancer, using bioinformatics technology to analyze the correlation between SKA1 and E2F1 expression, further detecting the binding site of E2F1 in the SKA1 promoter region, and using dual luciferase technology to detect E2F1 against SKA1. Transcriptional activation.Results:KA1 was highly expressed in liver cancer tissues, and the overall survival rate of liver cancer patients with high SKA1 expression was 49.8%, lower than that of patients with low SKA1 expression, showing a negative correlation. E2F1 is also highly expressed in liver cancer tissues, and the survival time of patients with liver cancer with high E2F1 expression is significantly lower than that in the low expression group, which was negatively correlated with poor prognosis. SKA1 overexpression could increase the proliferation ability of liver cancer cells by nearly 50%. SKA1 is regulated by the E2F1 transcription factor, and the E2F1 transcription factor is combined with the SKA1 promoter to transcriptionally activate the expression of SKA1 in liver cancer cells.Conclusion:E2F1 transcriptional activation of SKA1 promotes proliferation of hepatoma cells, leading to poor prognosis in patients with liver cancer

ABSTRACT

Objective: To determine the expressions of Versican and a distintegrin and metalloprotease with thrombospondin type l motifs (ADAMTS-1) in serum in the polycystic ovary syndrome (PCOS) patients, and to clarify their roles in the pathogenesis of PCOS. Methods: A total of 80 patients with PCOS ( PCOS group) and 100 healthy women (control group) were selected. The heights and body weights of the subjects in two groups were measured; and the body mass index (BMI) was calculated. The levels of serum Versican and ADAMTS-1 of the subjects in two groups were measured by enzyme-linked immunosorbent assay (ELISA) method. The levels of serum follicle stimulating hormone (FSH), luteinizing hormone (LH), testosterone (T), fasting blood glucose (FBG), fasting insulin of the subjects in two groups were detected; insulin resistance was evaluated according to Homeostasis Model Assessment of Insulin Resistance (HOMA-IR). The correlations between Versican, ADAMTS-1 and the metabolic indexes were analyzed by Pearson linear correlation analysis. Results: The serum Versican level of the patients in PCOS group was significantly decreased compared with control group (P=0.004); the serum ADAMTS-1 level was also decreased (P<0.01). The sensitivities, the specificities and the area under receiver operating characteristic (ROC) curve (AUC)of Versican and ADAMTS-1 in prediction of PCOS were 76.74%?vs 63.64% (P=0.018), 52.94% vs 40.73% (P=0.009) and 0675 (0.550-0.795) vs 0.714 (0.601-0.827) (P=0.032), respectively. The correlation analysis showed that in PCOS group the serum level of Versican of the patients was negatively correlated with FBG (r=-0.738, P=0.022), and ADAMTS-1 was negatively correlated with FBG (r=-0.524, P=0.043). Conclusion; Versican and ADAMTS-1 lowly express in the patients with PCOS. They are negatively correlated with the insulin resistance. They may play inhibitory effects in the occurrence of PCOS.

Fifteen patients were treated with tension band fixation (Group B) including 9 males and 6 females, with a mean age of (35.6 +/- 5.1) years. The indexes of intra-operation and during the hospital stay, the clinical outcomes and postoperative complications were statistically compared.

RESULTSAll the patients were followed up for 12 to 37 months, with an average of 20 months. There was no significant differences in data of operative time, blood loss and length of incision between the two group (P > 0.05); however, the economic cost was significantly much more in Group A (P < 0.01). Also, there was statistically difference in the incidence of complications between the two groups (P = 0.023), which were 2 patients in Group A and 8 patients in Group B. According to Rockwood evaluation criteria, in group A, 13 patients got an excellent result, 2 good and 1 poor; in group B, 10 patients got an excellent result, 3 good, 1 poor and 1 bad. There were no statistical difference in good and excellent rate.

CONCLUSIONThe clavicular hook plate for the treatment of Allman type II to III sternoclavicular joint dislocation is believed to be as good as tension band fixation, but there are fewer complications and in favor of early exercise.

ABSTRACT

The near infrared (NIR) spectroscopy technique has been applied in many fields because of its advantages of simple preparation, fast response, and non-destructiveness. We investigated the potential of NIR spectroscopy in diffuse reflectance mode for determining the soluble solid content (SSC) and acidity (pH) of intact loquats. Two cultivars of loquats (Dahongpao and Jiajiaozhong) harvested from two orchards (Tangxi and Chun'an, Zhejiang, China) were used for the measurement of NIR spectra between 800 and 2500 nm. A total of 400 loquats (100 samples of each cultivar from each orchard) were used in this study. Relationships between NIR spectra and SSC and acidity of loquats were evaluated using partial least square (PLS) method. Spectra preprocessing options included the first and second derivatives, multiple scatter correction (MSC), and the standard normal variate (SNV). Three separate spectral windows identified as full NIR (800approximately2500 nm), short NIR (800approximately1100 nm), and long NIR (1100approximately2500 nm) were studied in factorial combination with the preprocessing options. The models gave relatively good predictions of the SSC of loquats, with root mean square error of prediction (RMSEP) values of 1.21, 1.00, 0.965, and 1.16 degrees Brix for Tangxi-Dahongpao, Tangxi-Jiajiaozhong, Chun'an-Dahongpao, and Chun'an-Jiajiaozhong, respectively. The acidity prediction was not satisfactory, with the RMSEP of 0.382, 0.194, 0.388, and 0.361 for the above four loquats, respectively. The results indicate that NIR diffuse reflectance spectroscopy can be used to predict the SSC and acidity of loquat fruit.

ABSTRACT

The use of visible-near infrared (NIR) spectroscopy was explored as a tool to discriminate two new tomato plant varieties in China (Zheza205 and Zheza207). In this study, 82 top-canopy leaves of Zheza205 and 86 top-canopy leaves of Zheza207 were measured in visible-NIR reflectance mode. Discriminant models were developed using principal component analysis (PCA), discriminant analysis (DA), and discriminant partial least squares (DPLS) regression methods. After outliers detection, the samples were randomly split into two sets, one used as a calibration set (n=82) and the remaining samples as a validation set (n=82). When predicting the variety of the samples in validation set, the classification correctness of the DPLS model after optimizing spectral pretreatment was up to 93%. The DPLS model with raw spectra after multiplicative scatter correction and Savitzky-Golay filter smoothing pretreatments had the best satisfactory calibration and prediction abilities (correlation coefficient of calibration (R(c))=0.920, root mean square errors of calibration=0.196, and root mean square errors of prediction=0.216). The results show that visible-NIR spectroscopy might be a suitable alternative tool to discriminate tomato plant varieties on-site.

ABSTRACT

The use of near infrared (NIR) spectroscopy was proved to be a useful tool for quality analysis of fruits. A bifurcated fiber type NIR spectrometer, with a detection range of 800~2500 nm by InGaAs detector, was used to evaluate the firmness of peaches. Anisotropy of NIR spectra and firmness of peaches in relation to detecting positions of different parts (including three latitudes and three longitudes) were investigated. Both spectra absorbency and firmness of peach were influenced by longitudes (i, ii, iii) and latitudes (A, B, C). For modeling, two thirds of the samples were used as the calibration set and the remaining one third were used as the validation or prediction set. Partial least square regression (PLSR) models for different longitude and latitude spectra and for the whole fruit show that collecting several NIR spectra from different longitudes and latitudes of a fruit for NIR calibration modeling can improve the modeling performance. In addition, proper spectra pretreatments like scattering correction or derivative also can enhance the modeling performance. The best results obtained in this study were from the holistic model with multiplicative scattering correction (MSC) pretreatment, with correlation coefficient of cross-validation r(cv)=0.864, root mean square error of cross-validation RMSECV=6.71 N, correlation coefficient of calibration r=0.948, root mean square error of calibration RMSEC=4.21 N and root mean square error of prediction RMSEP=5.42 N. The results of this study are useful for further research and application that when applying NIR spectroscopy for objectives with anisotropic differences, spectra and quality indices are necessarily measured from several parts of each object to improve the modeling performance.
